Nearly all of the energy expended in human physiology is produced through what is known as the Krebs cycle (also known as the citric acid cycle), a process that takes place in the mitochondria, the microscopic “power plants” responsible for generating cellular energy. The cycle is an eightstep process that begins when acetyl-CoA—derived from the metabolism of carbohydrates, fats and amino acids—is bound to oxaloacetate to make citric acid (citrate). In the succeeding steps, citrate is converted, in turn, into the intermediaries isocitrate, alpha-ketoglutarate, succinyl coenzyme A, succinate, fumarate and malate, after which it is reconverted into oxaloacetate and the cycle begins again. During the course of the cycle, each molecule of citrate generates 36 molecules of ATP for tissue fuel.

The Importance of Krebs Intermediaries

While acetyl-CoA is the only fuel to enter the Krebs cycle, Krebs intermediaries may enter the cycle at many points. In fact, the cycle is primarily controlled by the availability of these Krebs intermediaries. In the normal function of the cycle, the intermediates are continually recycled through what are known as anaplerotic reactions to maintain constant concentrations in the mitochondria. However, many other biosynthetic processes also use these molecules as substrates, decreasing their availability for cellular respiration in what are known as cataplerotic reactions. Increasing the supply of any of these intermediaries can increase all of them as they are converted from one to another, ultimately increasing the ATP available to the cell.
